Running on uneven natural ground with consistent stability demanded skilled control, a factor crucial for human evolution. Circumnavigating hazardous obstacles, including steep drops, runners are further challenged by uneven ground, which, although less severe in nature, remains destabilizing. We are still uncertain about how foot placement is determined on irregular terrain and the implications for stability. In this respect, we monitored the energetics, kinematics, ground forces, and stepping patterns of human runners while traversing trail-like undulating uneven terrain. We have found that running steps are not focused on areas of consistent flatness. On the contrary, the body's automatic reaction, facilitated by leg flexibility, sustains balance without demanding precise footfall control. Moreover, their overall movement patterns and energy expenditure on rough surfaces demonstrated minimal variation compared to smooth surfaces. It is possible that these observations explain the means by which runners maintain stability across natural terrain while directing attention to tasks separate from the act of foot placement.

Singular enteroendocrine cells, however, commonly synthesize several, occasionally conflicting, gut hormones simultaneously; moreover, particular gut hormones are also manufactured in non-intestinal tissues. To selectively access enteroendocrine cells within mice, we developed in vivo strategies employing intersectional genetics. Within Vil1-p2a-FlpO knock-in mice, we strategically placed FlpO expression at the endogenous Villin1 locus, thereby ensuring reporter expression was limited to the intestinal epithelium. A range of skeletal dysplasias are a consequence of mutations occurring in the TRPV4 ion channel. In contrast, the mechanisms by which TRPV4 mutations influence the intensity of the disease remain unclear. Human-induced pluripotent stem cells (hiPSCs), genetically modified with CRISPR-Cas9 to harbor either the milder V620I or the more severe T89I mutations, were used to discern the differential impacts on channel function and chondrogenic differentiation. We observed enhanced basal currents through TRPV4 in hiPSC-derived chondrocytes, which were characterized by the V620I mutation. The mutations prompted an increased calcium signaling rate in response to the TRPV4 agonist GSK1016790A; however, the overall signal strength was diminished in comparison to the wild-type (WT). No differences were observed in the overall production of cartilaginous matrix, but the V620I mutation ultimately lowered the mechanical properties of the cartilage matrix at later stages of chondrogenesis. mRNA sequencing of the mutations revealed an upregulation of multiple anterior HOX genes and a downregulation of the antioxidant genes CAT and GSTA1 throughout the chondrogenesis process. BMP4's effect on wild-type chondrocytes was to upregulate several critical hypertrophic genes; yet, this hypertrophic maturation response was blocked in the mutant chondrocytes.
